SDS Hyb. Wash
Buffer #1, 1 liter
- 50 g of "cheap" SDS
- Add ~700mls H2O place on hot plate/stirrer, when "in" solution allow to cool while stirring then add :
- 40 mls of 2X NaHPO4 pH 7.2 = (134g of Na2HPO4.7H2O + 4 mls 85% H3PO4 / Liter)
- 2 mls of 0.5 M EDTA
- 16.6 mls 30% BSA; bring up to 1 liter pre-heat at 65oC
- WASH 2X's 15 minutes each at 65oC using ~500mls in a medium Tupperware container
Buffer #2, 1 liter
- 50 mls of 20% "cheap" SDS or 10g SDS
- 40 mls of 2X NaHPO4 pH 7.2 = (134g of Na2HPO4.7H2O + 4 mls 85% H3PO4 / Liter)
- 2 mls of 0.5 M EDTA pre-heat at 65oC
- WASH 2X's ~5-10 minutes each at 65oC (check by hand monitor)
- seal in Saran Wrap®
