Single Tube Submission
Please follow these guidelines when preparing your samples:
Each sample tube (1.5ml tubes preferred) should contain one primer and one template at the amount listed on the Order Form:
Please note that the values listed for the primer are quantities, not concentrations.
- 0.25μg DNA/5kb single stranded plasmid + 2 pmol primer in 13µl
- 0.5μg DNA/5kb double stranded plasmid + 3.2 pmol primer in 13µl
- 100ng DNA/1000bp PCR product + 0.8 pmol primer in 13 µl
If you need us to add the primer, put a large note in red on your order form, and your samples should be just 10uL total volume.
Template: Please be sure that you do not have SALTS, PROTEIN, DETERGENTS, RNA, CHROMOSOMAL DNA, ETHANOL, or EDTA
Quantitate your PCR products on an agarose gel or use a NanoDrop.
Primers: Please make sure your primers have a melting temperature (Tm) between 52°C and 60°C
